  • Prucalopride is a selective 5-HT4 receptor agonist that stimulates colonic peristalsis to increase bowel motility.
  • Suicides, suicide attempts, and suicidal ideation have been reported in clinical trials; a causal link to prucalopride has not been established, but patients should stop the medicine and contact their provider if depression worsens or suicidal thoughts emerge.
  • A history of hypersensitivity to prucalopride is a contraindication; reactions including rash, urticaria, and facial swelling have been observed.

How does Prucalopride work?

At therapeutic doses, prucalopride is selective for its target receptor and does not activate the other serotonin or gut-hormone receptors. More than 90% of an oral dose reaches the bloodstream, and the kidneys clear most of it, so the label warns against use in end-stage renal disease requiring dialysis and notes that a lower dose is needed in severe kidney impairment.
